Town, the highest ranked team in this season's competition, went ahead after five minutes when the prolific Emily Heckler stroked the ball past the advancing Bryony Hudson.
That was Town's first real foray into the Barnsley half and against the run of early play. Heckler was close to a second several minutes later, but fired a shot straight at Hudson from inside the six yard box.
Barnsley levelled ten minutes before half time with a fine strike from the left angle of the area by Amy Woodruff.
Barnsley had plenty of play in the second half and looked the more likely to get the game's third goal, but the deadlock remained unbroken.
Town came more into proceedings in the game's final quarter and with just under five minutes left, Hannah Spreckley headed the winner.
For Charley Evans, it was a second successive winning Final. She had been a key player when Barnsley had caused an upset a year ago and she, along with Emily Heckler (Doncaster Rovers Belles) and Carla Baxby (Rotherham United), joins those players who have won the trophy with different teams.
